有直接的影响,但是可上调颗粒细胞中CYP2A4 和CYP19A1 的表达,进而促进颗粒细胞中E2 和孕酮的
分泌,促进卵巢的发育[19-20]。本试验发现高浓度褪黑素的添加在24h时可以显著增强BMP6 的表达,之
后会呈现抑制的现象可能是存在机体自身的负反馈调节,72h后,高浓度(10-3 mol/L)和低浓度褪黑素
(10-11 mol/L)对BMP6 的表达没有影响,其他浓度均显著促进其表达。
本研究验证了褪黑素对于卵巢颗粒细胞发育相关基因的影响,结果表明,不同浓度的褪黑素可以通过
促进抗氧化相关基因(GPX4、SOD1)、发育相关基因(EGFR、DNMT1a)和TGFβ家族(BMP6)基因的表
达,来减少氧化应激对颗粒细胞功能的影响,刺激颗粒细胞的发育成熟,同时抑制FSHR 的表达,保护颗
粒细胞抑制其凋亡。
