分子生物学试剂市场基本上都是提供野生
型的 DNA 聚合酶。随着客户实验要求的提
升和生物样本复杂性的提高,野生型 DNA
聚合酶的局限性在许多实验中也逐渐凸显
出来,如扩增效率低,特异性差,高 AT 或
GC 含量样本扩增结果差,样本中或 PCR 系
统中有些抑制剂对反应有干扰等等。
自 2017 年 1 月 1 日起,默克成为罗氏旗
下 KAPA Biosystems 产品的独家供应商,
在美国、巴西和日本以外的所有国家提供聚
合酶链反应 (PCR) 和实时定量聚合酶链反
应(qPCR)所用的全新酶及试剂盒产品。
KAPA Biosystems 的 PCR 和 qPCR 产品
在实验中的优异表现主要源自它提供的
DNA 聚合酶是通过 Directed Evolution
工程化修饰的。野生型 DNA 聚合酶基因经
过多轮的突变,筛选和扩增,最终获得客户
需要的特性,如增加酶的合成能力或降低酶

DNA Polymerase
Total PCR times for the generation of different sized amplicons for
human genomic DNA (≤3.5 kb) and lambda (≤5 kb) using wild-type
Taq DNA polymerase and KAPA2G Fast DNA Polymerase.
The amount of time that can be saved using protocols based on wild-type
Taq is limited by the extension rate of the enzyme. KAPA2G Fast DNA Polymerase is based on a secondgeneration polymerase with an ability to synthesize DNA faster than wild-type Taq or other DNA polymerases. Reaction
times are based on a 35-cycle program using the cycling profile recommended by each kit manufacturer.

Amplification of 5 human gene fragments using KAPA2G Fast HotStart or competitor hot-start Taq formulations.
Reactions (25 μL) contained 5 ng human genomic DNA and 0.5 units (KAPA2G Fast HotStart and Competitor I) or 0.625 units (Competitor A and Q)
enzyme. For amplicons with a GC content >65% (lanes 2 and 3), 7.5%
DMSO was included in reactions. A 3-step cycling profile (35 cycles) with 15
sec denaturation (95˚C) and 15 sec annealing (60˚C) per cycles was used
for all enzymes. The extension (72˚C) was 1 sec/cycle for KAPA2G Fast HotStart and 60 sec/cycle for competitor enzymes. The total reaction time for
each enzyme is indicated.

No bias in amplification efficiency across a wide range of GC
contents (44.2 – 62.5%) or amplicon lengths (86 – 249 bp)
was observed with KAPA SYBR FAST.
Amplification efficiencies achieved for ten housekeeping genes with
the KAPA SYBR FAST Universal qPCR Kit were plotted against GC content (left) or amplicon length (right). The reaction efficiency achieved
for each of the ten genes fell within the optimal range of 95 – 105%,
independent of the nature or length of the amplicon.
Two target genes were amplified using a
10-fold dilution series of human genomic DNA
(16 ng to 1.6 pg per 20 μL reaction) comparing four commercial kits. KAPA SYBR FAST
